چکیده
The pathogenesis of the myopathy occurring in the heart of the cardiomyopathic strain of the Syrian hamster is not well understood but is believed to be associated with abnormal calcium handling by myopathic cells. The purpose of this study was to determine whether the cardiomyopathy occurring in strain BIO 14.6 animals is associated with an enhanced a1-adrenergic receptor-mediated rise in cytosolic calcium, whether a pertussis toxin-sensitive G protein is involved in coupling the al-adrenergic receptor to changes in intracellular calcium and whether enhanced al responsiveness is associated with an increase in the level of expression of the a1-adrenergic receptor or in the pertussis toxin-sensitive G protein or proteins. To test the hypothesis that the cardiomyopathic state is associated with a greater ael-receptor-mediated rise in cytosolic calcium, we studied the effect of phenylephrine (in the presence of propranolol) on time-averaged cytosolic calcium concentration ([Ca2+] ) in isolated cardiac myocytes from cardiomyopathic and age-matched control hamsters. Phenylephrine caused a greater increase both in time-averaged [Ca2"ji (an increase of 48±8% versus 12 ±3%, p <0.01) and in contractility (+ 181±22% versus +35+9%, p <0.01) in cardiomyopathic than in normal cardiac myocytes. Exposure to pertussis toxin (200 ng/ml for 3 hours) attenuated the cc1-adrenergic receptor-mediated increase in contractility and time-averaged [Ca2+]i in both cardiomyopathic and normal cells. The level of pertussis toxin-sensitive G protein, as determined by pertussis toxin-mediated [32P]A)DP-ribosylation, was 1.6-fold higher in cardiomyopathic versus normal hamster hearts. The density of a1-adrenergic receptors, as measured by the antagonist radioligand [3Hlprazosin and the affinity of the receptor for agonist and antagonist were similar in myopathic and normal heart membranes. Thus, in cardiac myocytes from hamsters, the al-adrenergic receptor-mediated effects on [Ca2`]i and contractility appear to be mediated by a pertussis toxin-sensitive G protein or proteins. In myocytes from cardiomyopathic hamsters, these al-adrenergic effects were increased in magnitude, as was the level of pertussis toxin-sensitive G protein, but there was no measurable alteration in the density or ligand binding properties of al-adrenergic receptors. Effect of imipramine and desipramine on adenosine receptors in isolated rat atria
The effect of different doses (1-50 µ M) of imipramine (IMI) and desipramine (DES) on the rate and force of contraction of isolated rat atria was studied. IMI and DES produced a dose-dependent increase in force of contraction (31- 94% for IMI and 35-118% for DES). Pretreatment of rats with reserpine (5 mg/kg) on the isolated atria with propranolol (1 µ g) inhibited the positive ionotropic eff...
متن کاملThe Role of Adrenergic Receptors on Neural Excitability and Synaptic Plasticity: A Narrative Review
Adrenergic receptors have an important role in neural excitability and synaptic plasticity. Despite a lot of studies on these receptors, their exact role in brain disorders accompanied with hyperexcitability has not been determined. There are also controversies on their role in synaptic plasticity. In this review article, the important studies done in this regard have been reviewed to achieve a...
متن کاملa1-Adrenergic Receptors Function Within Hetero-Oligomeric Complexes With Atypical Chemokine Receptor 3 and Chemokine (C-X-C motif) Receptor 4 in Vascular Smooth Muscle Cells
Background-—Recently, we provided evidence that a1-adrenergic receptors (ARs) in vascular smooth muscle are regulated by chemokine (C-X-C motif) receptor (CXCR) 4 and atypical chemokine receptor 3 (ACKR3). While we showed that CXCR4 controls a1ARs through formation of heteromeric receptor complexes in human vascular smooth muscle cells (hVSMCs), the molecular basis underlying cross-talk between...
متن کاملIn vivo Characterization of Fusion Protein Comprising of A1 Subunit of Shiga Toxin and Human GM-CSF: Assessment of Its Immunogenicity and Toxicity
Background: Most cancer cells become resistant to anti-cancer agents. In the last few years, a new approach for targeted therapy of human cancer has been developed using immunotoxins which comprise both the cell targeting and the cell killing moieties. Methods: In the present study, the recombinant Shiga toxin A1 subunit fused to human granulocyte-macrophage colony stimulating factor (A1-GM-CSF...
متن کاملa1-Adrenergic Receptor Subtypes in Human Peripheral Blood Lymphocytes
We investigated the expression of a1-adrenergic receptor subtypes in intact human peripheral blood lymphocytes using reverse transcription–polymerase chain reaction (RT-PCR) and radioligand binding assay techniques combined with antibodies against the three subtypes of a1-adrenergic receptors (a1A, a1B, and a1D). RT-PCR amplified in peripheral blood lymphocytes a 348-bp a1A-adrenergic receptor ...
